Mammalian cell morphology is a key determinant of function, igniting significant interest in understanding how cell phenotype is regulated by microenvironmental cues. These cues include cell-cell interactions, soluble signaling molecules, and adhesion to the extracellular matrix (ECM) via transmembrane receptors; the physical properties of the ECM, particularly its stiffness and rate of stress relaxation are known to regulate the ability of a variety of cell types to spread and differentiate in 2D and 3D culture. However, many of these properties are interconnected and developing material systems to isolate individual properties becomes an important tool; a tool that allows for the investigation of the changes ECM materials undergo to better understanding disease states and the therapeutic opportunities. My laboratory will tackle these issues by synthesizing novel polymeric architectures to study prenatal membrane rupture, keratinocyte stem cell differentiation and callus formation, the timescales of immune cell mechanotransduction, and developing new tools to measure in situ mechanical forces in viscoelastic materials. My training in materials science has me uniquely positioned to not only synthesize new tools and methods to study ECM interactions, but in adapting our findings and working with interdisciplinary teams to apply them for therapeutic and commercial benefit.
